Fraunhofer vs. Corona: Drug Repurposing Using the Example of Inhalation Therapy Against Covid-19

New drugs against Covid-19 are urgently needed. Repurposing of existing drugs enables faster development and minimizes risks. Inhalable drugs offer the benefits of non-invasive administration and less systemic side effects. Within the DRECOR project, we selected and identified drug repurposing candidates using in-vitro/in-vivo inhalation studies and PK/Tox assessments.
PBPK Modeling: The Road to Systemic Toxicity from External to Internal Exposure

Physiologically-based pharmacokinetic (PBPK) models intrinsically link external and internal exposure, dose response, and target organ toxicity. Integration of innovative in vitro test systems and knowledge on in vivo physiological parameters are crucial for an integrated approach to testing and assessment (IATA). Our PBPK model specifically addresses the complex dosimetry for inhalation toxicity.

Nowadays, people spend most of their lives indoors – at work, home, schools, or traveling. Indoor air pollution has become an increasingly important issue, with one focus on ventilation and filter systems. Fraunhofer ITEM combines expertise in inhalation toxicology with chemical analyses and human exposure for selected scenarios.
